'Isolation' is a question of 'collision domains' and broadcast traffic.
Multiple NICs on the same subnet will increase the load
regardless of protocol (it's all Ethernet frames @ that level).
Broadcast packets are received and processed by the CPU
and can not be filtered via MAC address.

Only direct cabling eliminates both of those problems.
Subnet masks are only good for TCP/IP and eliminates neither
problem as the mask is applied only to determine routing out
of the NIC. Protocol bindings/different protocols eliminate neither
problem as all protocols have some broadcast traffic and they
are still on the same wire. Workgroup names only work at the
Microsoft Client (OSI application) layer.
