You can measure backlash with a dial indicator. Set it up to measure table movement in the direction you are working with. Turn the screw in one direction (slowly if manual, or .001 jog). Bring the indicator to a known spot (like zero). Advance the screw in the other direction until the table just starts to move. The amount it should have moved minus what it did move is backlash.
How much is acceptable is up to you and the tolerance of the work you do.
